Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Pre-mRNA processing factor involved in disassembly of spliceosomes after the release of mature mRNA.
Gene References into Functions
  1. These data indicate that the U2 snRNP-intron interaction is Prp43's major target. PMID: 27115347
  2. Using Prp43 F357A mutants or pyrimidine nucleotides, we show that the lack of stacking of the nucleotide base to the F-motif decouples the NTPase and helicase activities of Prp43. In contrast the R159A mutant (R-motif) showed reduced ATPase and helicase activities. PMID: 28180308
  3. Study shows the crystal structure of Prp43p bound to single-stranded poly-uracil RNA and reveals differences with viral ATPases in the RecA1 domain but with striking similarities with viral ATPases in the RecA2 domain of the catalytic core, including both a prominent beta-hairpin that bookends the 5' end of the RNA stack and a dynamic motif Va that is implicated in mediating translocation. PMID: 28416566
  4. Results show that Prp43 expression is regulated by Cmg1 which stimulates its RNA helicase activity. PMID: 26821976
  5. Prp43 differentially interacts with g-patch domains in Spp382, Sqs1, and Pxr1 G. PMID: 25808954
  6. This study investigated the activation of Prp43 by Ntr1 in the presence and absence of RNA substrate. PMID: 24165877
  7. The DEAH box ATPases Prp16 and Prp43 cooperate to proofread 5' splice site cleavage during pre-mRNA splicing. PMID: 20705241
  8. Prp43p promotes the discard of suboptimal and optimal 5' exon and lariat intermediates indiscriminately. PMID: 20463285
  9. Data report that cleavage of pre-rRNA at the 3'-end of 18 S rRNA, requires functional interactions between Nob1 and Prp43, and the cofactor Pfa1. PMID: 19801658
  10. stimulation of ATPase/helicase activities of Prp43p by Pfa1p is required for efficient 20S pre-rRNA-to-18S rRNA conversion PMID: 19927118
  11. The ATPase activity of Prp43p is required for early steps of pre-rRNA processing and normal accumulation of mature rRNAs. PMID: 16227579
  12. Prp43p promotes recycling of snoRNAs and biogenesis factors during pre-rRNA processing. The dual function for Prp43p in the cell raises the possibility that ribosome biogenesis and pre-mRNA splicing may be coordinately regulated. PMID: 16382143
  13. Its essential function is in its newly characterized role in ribosome biogenesis of both ribosomal subunits, positioning Prp43p to regulate both pre-mRNA splicing and ribosome biogenesis. PMID: 16382144
  14. We report that Prp43 hydrolyzes all common NTPs and dNTPs and unwinds short 5'/3' tailed RNA/DNA duplexes in an ATP-dependent fashion. PMID: 16700561
  15. Prp43 acts on preribosomal regions surrounding each binding site, with distinct functions at different locations. PMID: 19941819
