Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Forms the histone H2A phosphatase complex in association with the regulatory subunits PSY2 and PSY4, which dephosphorylates H2AS128ph (gamma-H2A) that has been displaced from sites of DNA lesions in the double-stranded DNA break repair process. Dephosphorylation is necessary for efficient recovery from the DNA damage checkpoint. PPH3 is directly involved in the dephosphorylation and activation of the transcription factor GLN3 in response to nutrient availability.
Gene References into Functions
  1. Physical interaction between Pph3 and Mec1, mediated by cofactors Psy2 and Ddc2, is important for regulating the checkpoint response in yeast. PMID: 25533186
  2. Data indicate that the phosphatase complex Pph3/Psy2 connects homologous end-joining (NHEJ) repair to cell cycle progression. PMID: 24498054
  3. Data show that Pph3-Psy2 targets the glucose signal transducer protein Mth1, which is required for the dephosphorylation of Rgt1 upon glucose withdrawal, a critical event in the repression of HXT genes, which encode glucose transporters. PMID: 24277933
  4. a three-protein complex (HTP-C, for histone H2A phosphatase complex) containing the phosphatase Pph3 that regulates the phosphorylation status of gammaH2AX in vivo and efficiently dephosphorylates gammaH2AX in vitro PMID: 16299494
  5. Mutants lacking either the phosphatase activator Rrd1 or the phosphatase Pph3 are resistant to rapamycin, double mutants exhibit a synergistic response. PMID: 16322523
  6. Rad53 regulates replication fork restart and initiation of late firing origins independently PMID: 17517611
  7. Mutational analyses of conserved regions of Tap42/alpha4 identified a region critical for its interactions with PP2Ac. We propose a scaffolding function for Tap42/alpha4 whereby the interaction of PP2Ac promotes the dephosphorylation of substrates. PMID: 17616149
  8. Results suggest that the trimeric Pph3p-Psy4p-Psy2p complex may dephosphorylate both gammaH2AX and Rad53p. PMID: 18647348
