Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Required for anaphase spindle elongation and microtubule bundling in both interphase and mitosis. Has a role in spatial and temporal regulation of septation and cytokinesis and ensures equal partition of segregating sister chromatids. Ensures correct midzone positioning of protein kinase ark1. Acts as a regulatory component at cytokinesis checkpoint where it inhibits nuclear division when actomyosin ring formation is impaired.
Gene References into Functions
  1. Study shows that fission yeast bipolar spindle assembly and chromosome segregation can take place in the absence of both kinesin-5 Cut7 and kinesin-14 Pkl1. In cut7Deltapkl1Delta cells, spindle formation and bipolarity requires the microtubule (MT) antiparallel bundler Ase1 to recruit Cls1 to stabilize MTs. PMID: 28513584
  2. Study demonstrated that diffusible microtubule crosslinker Ase1 generated directed microtubule sliding when confined between partially overlapping microtubules. The Ase1-generated forces, directly measured by optical tweezers to be in the piconewton-range, were sufficient to antagonize motor-protein driven microtubule sliding. PMID: 25748652
  3. Show that Ase1, a member of the conserved MAP65/PRC1 family of microtubule-bundling proteins, enables the formation of stable antiparallel overlaps through adaptive braking of Kinesin-14-driven microtubule-microtubule sliding. PMID: 21892183
  4. An Ase1/Prc1-dependent mechanism that prevents spindle collapse and thus asymmetric division is required for correction of merotelic attachment in anaphase. PMID: 19948483
  5. ase1 has a role in mitotic cell division, meiotic nuclear oscillation, and cytokinesis checkpoint signaling PMID: 15647375
  6. The ase1 protein functions to organize microtubules into overlapping antiparallel bundles both in interphase and mitosis. This dual function suggests that ase1 is differentially regulated through the cell cycle. [ase1] PMID: 15689489
  7. loss of Ase1 activates a checkpoint that requires only the Mad3, Bub1, and Mph1, but not Mad1, Mad2, or Bub3 checkpoint proteins PMID: 18562692
  8. Cells use a finely tuned cooperative localization mechanism that exploits differences in the geometry and concentration of ase1p binding sites along single and overlapping microtubules. PMID: 18976915
  9. Data show that fission yeast kinesin-6 motor klp9p binds to the microtubule antiparallel bundler ase1p at the midzone at anaphase B onset. PMID: 19686686
