  • IHC image of CSB-RA080928A0HU diluted at 1:100 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.

Description

VAMP1, also known as Synaptobrevin-1, serves as a critical component of the SNARE complex that mediates synaptic vesicle fusion with the presynaptic membrane. This vesicle-associated membrane protein plays an essential role in neurotransmitter release, making it a key target for researchers investigating synaptic transmission, neuronal communication, and related neurological disorders.

This recombinant monoclonal antibody (clone 25H6) offers significant advantages for neuroscience research through its sequence-defined production in rabbit host. Unlike traditional hybridoma-derived antibodies, recombinant technology ensures consistent performance across experiments and eliminates the lot-to-lot variability that can compromise longitudinal studies or multi-site collaborations. The defined sequence also provides a stable, renewable resource for ongoing research programs.

Validation in immunohistochemistry demonstrates reliable detection of VAMP1 in paraffin-embedded human brain tissue, with recommended working dilutions of 1:50 to 1:200. The validated protocol employed citrate buffer antigen retrieval at pH 6.0 under high pressure conditions, followed by overnight primary antibody incubation at 4°C and detection via HRP-conjugated goat anti-rabbit polymer system with DAB visualization. This established workflow provides researchers with a proven starting point for their own tissue studies. The antibody is also validated for ELISA applications, offering flexibility for both quantitative and spatial analysis approaches.

Supplied in a glycerol-containing buffer optimized for long-term stability at -20°C or -80°C, this affinity-purified antibody is well-suited for investigations into synaptic function, vesicle trafficking mechanisms, and neurodegenerative conditions where SNARE-mediated processes may be disrupted.
