Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4℃ for up to one week.

Target Background

Function
Outer membrane channel, which is required for the function of several efflux systems such as AcrAB-TolC, AcrEF-TolC, EmrAB-TolC and MacAB-TolC. These systems are involved in export of antibiotics and other toxic compounds from the cell. TolC is also involved in import of colicin E1 into the cells.
Gene References into Functions
  1. TolC, a potentially interesting pharmaceutical target can be addressed by small molecules, blocking the channel. Biophysical characterization of the binding processes will support future identification and optimisation of more potent TolC blockers in order to validate TolC as a pharmaceutical target. PMID: 28746830
  2. The authors report the near-atomic resolution cryoEM structures of the Escherichia coli AcrAB-TolC multidrug efflux pump in resting and drug transport states, revealing a quaternary structural switch that allosterically couples and synchronizes initial ligand binding with channel opening. PMID: 28355133
  3. A number of multi-drug efflux genes, particularly the central component TolC, show higher expression in antibiotic resistant Escherichia coli. PMID: 27105118
  4. Data suggest that outer membrane channel TolC plays an important role in extrusion of isoprenol and other medium-chain alcohols (CmOHs). PMID: 26563610
  5. Thus, tolC mutants grown in low iron conditions accumulate periplasmic enterobactin, which impairs bacterial morphology, possibly by sequestering iron and inhibiting an iron-dependent reaction involved in cell division or peptidoglycan synthesis. PMID: 24330203
  6. Interactions between TolC and AcrAB are highly dynamic and only a small fraction of TolC is utilized to support efflux activities. PMID: 23331412
  7. The TolC enhancement of extreme-acid survival includes Gad induction, but TolC-dependent growth rates below pH 6.5 do not involve Gad. PMID: 21541325
  8. These results indirectly suggest that TolC and YiaT proteins may play some role(s) in multidrug resistance. PMID: 20639525
  9. Inactivation of TolC triggers metabolic shutdown in E. coli cells grown in minimal glucose medium. PMID: 20545840
  10. essential for enterobactin export across the outer membrane PMID: 16166532
  11. The mechanism of tetracycline resistance was studied using expression studies in E. coli mutants that were defective in the tet(A) and the tolC proteins. PMID: 16687460
  12. Dada show that a combination of sbmA and tolC mutations appears to have additive effect, resulting in almost complete suppression of the phenotypic expression of Tn10 tetracycline resistance. PMID: 18083810
  13. Results describe two crystal structures of the Escherichia coli outer membrane protein TolC in its partially open state. PMID: 18406332
  14. TolC-dependent efflux system is involved in the exclusion of porphyrins in E. coli. PMID: 18641137
  15. The authors conclude that TolC plays an important role in L: -cysteine tolerance probably due to its export ability and that TolC overexpression is effective for L-cysteine production in E. coli. PMID: 18828007
  16. Data that TolC is involved in the efflux of certain cellular metabolites that accumulate during growth and trigger the upregulation of MarA, SoxS, and Rob, which in turn upregulate tolC and help rid the bacteria of these metabolites. PMID: 19502391
