Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Notes

Description

To produce the recombinant human sphingomyelin phosphodiesterase 2 (SMPD2) in an in vitro E. coli expression system, the gene fragment encoding the full-length SMPD2 protein (1-423 aa) is cloned into a suitable expression vector with appropriate regulatory elements. Next, isolate and purify the recombinant plasmid DNA containing the cloned gene fragment. An in vitro transcription reaction is then set up using the purified plasmid DNA as a template, resulting in mRNA synthesis. Subsequently, an in vitro translation reaction is performed using the synthesized mRNA to allow protein synthesis. The recombinant human SMPD2 protein is purified from the reaction mixture. SDS-PAGE analysis confirms its purity, reaching up to 90%. The protein migrates as a band with a molecular weight of 44 kDa on the gel.

Catalyzes the hydrolysis of sphingomyelin to form ceramide and phosphocholine. Ceramide mediates numerous cellular functions, such as apoptosis and growth arrest, and is capable of regulating these 2 cellular events independently. Also hydrolyzes sphingosylphosphocholine. Hydrolyze 1-acyl-2-lyso-sn-glycero-3-phosphocholine (lyso-PC) and 1-O-alkyl-2-lyso-sn-glycero-3-phosphocholine (lyso-platelet-activating factor).
