Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
This is a receptor for VIP as well as PACAP-38 and -27, the activity of this receptor is mediated by G proteins which activate adenylyl cyclase. Can be coupled to phospholipase C.
Gene References into Functions
  1. Findings suggest that the vasoactive intestinal peptide receptor 2 plays an important role in the fear extinction processes and the regulation of the dendritic morphology in the prelimbic and infralimbic cortices. PMID: 29030297
  2. Results demonstrate critical protective roles for PACAP and the VPAC2 receptor against autoimmunity, promoting the expansion and maintenance of the Treg pool. PMID: 25305591
  3. The results suggest that caveolin-1 plays an important role in VPAC2 receptor internalization and desensitization. PMID: 23499767
  4. Cumulus cells express PACAP and its receptors PAC1-R and VPAC2-R. PMID: 23684890
  5. VPAC2R mRNA expression showed a nonsignificant tendency to increase on day 7 after ischemia induced by bilateral common carotid artery occlusion. PMID: 22669509
  6. The loss of adrenal clock gene rhythm in the VPAC2 receptor knockout mice after transfer into constant darkness was accompanied by disappearance of rhythmicity in Star mRNA expression and serum corticosterone concentration. PMID: 22622901
  7. Cyclophosphamide-induced cystitis significantly decreased VPAC2 receptor expression in the urothelium & detrusor of both WT and NGF-OE mice PMID: 22700375
  8. The circadian clock, not ambient light level, regulates Vipr2 protein localization, since VAPC2R plays a role in signaling at all times of day. PMID: 22684939
  9. while the main rhythm parameters of extra-SCN circadian oscillations are maintained in Vipr2(-/-) mice, the ability of these oscillators to resist phase shifts is compromised PMID: 21559484
  10. Cellular synchrony in electrical firing rates was lost in suprachiasmatic nucleus slices lacking the VPAC2 receptor for VIP. PMID: 21628548
  11. The absence of VPAC2 leads to aberrant antibody production in Aspergillus fumigatus sensitized and challenged mice PMID: 20923692
  12. VPAC2 signaling is necessary for a functional circadian clock driving locomotor activity, core body temperature, and heart rate rhythmicity. VPAC2-deficient mice lose the rhythms in all three parameters under constant light/darknessconditions. PMID: 21178124
  13. absence of VPAC(2) does not protect mice from developing the signs and symptoms of allergic asthma. PMID: 20226823
  14. We also show that VPAC(2) receptor activation shifts the fate of symmetrically dividing NSPCs toward a nestin-only phenotype, independent of the trophic effect. PMID: 19650041
  15. Mice lacking the VPAC2 receptor for VIP and PACAP lose normal circadian rhythms of rest/activity behavior. They also lose circadian expression of the clock genes mPer1, mPer2, mCry1 and arginine vasopressin in the SCN. PMID: 12086606
  16. RT PCR demonstrated mRNA for VPAC2 in the trigeminal ganglion. Immunocytochemistry showed VPAC2 in small-diameter TG neurons. Thus, PACAP and VIP are potential mediators of cross-excitation in the TG. PMID: 12225867
  17. VPAC2 receptor is found predominantly in smooth muscle (in blood vessels and in the smooth muscle layers of the gastrointestinal and reproductive systems), mucosal epithelium in the colon, lung, the vasculature of the kidney, adrenal medulla, and retina PMID: 14617572
  18. Absence of photic gating, together with the general lack of overt rhythms in circadian output, strongly suggests the circadian pacemaker is completely dysfunctional in the Vipr2(-/-) mouse PMID: 15071099
  19. Vasoactive intestinal peptide binding to VPAC(2) on CD4 T cells specifically induces an up-regulation of the Th2-type transcription factors c-Maf and JunB, which consequently enhances IL-4 and IL-5 production, leading to a Th2-type phenotype. PMID: 15187104
