Target Background

Function
Activator protein that regulates the ubiquitin ligase activity and substrate specificity of the anaphase promoting complex/cyclosome (APC/C). During telophase and in the subsequent G1 phase of the cell cycle, recognizes and binds proteins containing a destruction box (D-box) and an additional degradation signal termed the KEN box including ASE1, CDC20, the B-type cyclins CLB2 and CLB3, the polo-like kinase CDC5 and HSL1, and recruits them in a C-box-dependent manner to the APC/C for ubiquitination and subsequent proteolysis. Required for exit from mitosis, cytokinesis and formation of prereplicative complexes in G1. Probably is the target of a BUB2-dependent spindle checkpoint pathway.
Gene References into Functions
  1. results suggest that binding to the D-box receptor may be a shared requirement for recognition and processing of all Cdh1 substrates. PMID: 27226622
  2. This study showed that the APC/C(Cdh1) ubiquitinates Ndd1 and mediates its degradation, and that APC/C(Cdh1) activity suppresses accumulation of Ndd1 targets. PMID: 25959309
  3. Cdh1 mediates Cdc20 degradation in metaphase, promoting formation of the APC/C-Cdh1. PMID: 25025567
  4. the structure of an Acm1-Cdh1 complex PMID: 23707760
  5. Acm1 is not required to inhibit APC(Cdh1) activity but rather prevents untimely Cdh1-substrate interactions. PMID: 22189709
  6. Tight Cdh1 binding combined with the inaccessibility of ubiquitinatable lysines contributes to pseudosubstrate inhibition of anaphase promoting complex(Cdh1). PMID: 21460798
  7. Data show that different spindle-regulatory pathways have distinct sensitivities to Cdh1, and ACM1 may buffer essential CDK phosphorylation of Cdh1. PMID: 20089834
  8. phosphorylation of CDK sites promotes the subsequent recognition of Cdh1 by at least one additional kinase PMID: 15467459
  9. Data demonstrate that Cdh1p interacts directly with the D box and the KEN box, two motifs within anaphase-promoting complex (APC) substrates known to be required for APC-mediated degradation. PMID: 15916960
  10. Data imply that the anaphase-promoting complex/cyclosome (APC/C) is activated by association of Cdh1 with Cdc27, which enables APC/C to recognize the D box of substrates via Cdh1's propeller domain. PMID: 15916961
  11. As cells complete mitosis and disassemble the spindle, the ubiqutin ligase APC(Cdh1) targets Fin1 for destruction. PMID: 17173039
  12. Cdh1-mediated degradation of the Polo kinase Cdc5 promotes the return of Cdc14 into the nucleolus. PMID: 18172166
  13. Data show that in budding yeast the activation of APC(Cdh1) is controlled in part by destruction of the Cdh1 inhibitor Acm1. PMID: 18498748
  14. unique property of the Acm1 degron sequences results from an unusually high affinity interaction with the substrate receptor site on the WD40 domain of Cdh1 that may serve both to promote APC inhibition and protect Acm1 from destruction. PMID: 18596038
  15. a role of G(1) specific APC/C(Cdh1) activity is to reset substrate levels to enables appropriate regulation of substrate accumulation in the subsequent phases of the cell cycle. PMID: 19713762
