Description

The production of the griffithsin polyclonal antibody involves a stepwise process. Initially, a rabbit is subjected to multiple immunization rounds using a recombinant Griffithsia sp. griffithsin protein (1-121aa). Once a sufficient antibody titer is achieved, the rabbit's blood is collected, and the antibodies are purified from the serum via the use of protein G. The functionality of the resulting griffithsin antibody is subsequently evaluated in ELISA and WB applications, confirming its capability to specifically interact with the Griffithsia sp. griffithsin protein.

Griffithsin (GRFT), a homodimeric high-mannose oligosaccharide binding lectin sourced from the red alga Griffithsia sp., shows broad-spectrum antiviral activity against HIV at picomolar concentrations. Its antimicrobial efficacy stems from its capacity to interact with viral envelope glycoproteins, particularly targeting terminal mannoses on oligosaccharides found in various enveloped viruses, including HIV-1, HCV, and SARS-CoV.

Function
Mixed specificity lectin with anti-HIV activity. Binds to HIV envelope glycoproteins, including exterior membrane glycoprotein gp120, and inhibits viral entry into cells. Binding to gp120 is dependent on gp120 being glycosylated, and is inhibited by mannose, glucose and N-acetylglucosamine.
