Target Background

Function
Acts as component of the MCM2-7 complex (MCM complex) which is the putative replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The six ATPase active sites, however, are likely to contribute differentially to the complex helicase activity; specifically the MCM2-MCM5 association is proposed to be reversible and to mediate a open ring conformation which may facilitate DNA loading. Once loaded onto DNA, double hexamers can slide on dsDNA in the absence of ATPase activity.
Gene References into Functions
  1. The Mcm ATPase active sites that flank Mcm2/5 (i.e. Mcm5/3 and 6/2) modulate the biochemical activities of the putative Mcm2/5 gate. PMID: 20484375
  2. A domain-push model is proposed to explain the bypass activity of mutant proline62leucine BOB1, which has a major effect on helicase activity, but no significant impact on DNA binding or ATPase of Methanobacterium thermoautotrophicum MCM gene. PMID: 16401076
  3. Mcm5 protein has a positive role in origin binding, which requires coordination by all six Mcm2-7 subunits in the hexamer. PMID: 18660534
  4. Conclusions predict a structural discontinuity between Mcm2 and Mcm5 and demonstrate that in contrast to other hexameric helicases, the six Mcm2-7 active sites are functionally distinct. PMID: 18662997
  5. DNA runs through a central channel in the double hexamer, and, once loaded, Mcm2-7 can slide passively along double-stranded DNA. PMID: 19896182
