Description

To generate the nos polyclonal antibody, a recombinant Drosophila melanogaster (Fruit fly) nos protein (amino acids 211-401) is used as the immunogen to elicit an antibody response in a rabbit. The nos antibody is collected from the rabbit serum and then undergoes affinity chromatography purification. The functionality of the nos antibody is confirmed through ELISA and WB assays, validating its ability to detect the Drosophila melanogaster (Fruit fly) nos protein in experimental settings.

The Nanos (nos) protein in Drosophila melanogaster is a pivotal regulator of embryonic development. Acting as a maternal-effect gene, Nanos contributes to the specification of cell fates along the anterior-posterior axis of the embryo, with a specific emphasis on the posterior region. Its role extends to the formation and specification of primordial germ cells, essential for the development of reproductive cells in the adult organism. Nanos influences posterior morphogenesis, playing a key role in defining the body plan and contributing to the proper formation of the posterior pole of the embryo.

Target Background

Function
Maternal RNA-binding protein that is required for germ cells proliferation and self-renewal. Acts by forming a complex with pum and brat that regulates translation and mRNA stability. The complex binds to the Nanos Response Element (NRE), a 16 bp sequence in the hb mRNA 3'-UTR and prevents its translation. Controls posterior development. Rescuing factor for the abdominal defect of posterior group mutants. The other posterior group genes are not required for nanos function but rather play a role in localization or distribution of nanos protein.
Gene References into Functions
  1. Nos protein has dual functions in translational repression and stabilization of specific RNAs to ensure proper germline development. PMID: 29278271
  2. germ stem cell-rescuing activity of Wolbachia TomO protein appears to be tightly coupled to the ability to enhance Nos protein expression PMID: 29701280
  3. TomO enhances the maintenance of germ stem cells (GSCs) by elevating Nanos (Nos) expression via its interaction with nos mRNA, ultimately leading to the restoration of germ cell production in Sxl mutant females that are otherwise without GSCs.[ PMID: 27498563
  4. Translational repression of the Drosophila nanos mRNA involves the RNA helicase Belle and RNA coating by Me31B and Trailer hitch.Bel is required for nos mRNA translational repression. PMID: 28701521
  5. Results showed that nos mRNA, one of the target mRNAs of Hrp38 identified in this study, is regulated by Hrp38 for translational control during ovary development. PMID: 27402862
  6. The results of this experimentat showed that dynein motor machinery components mediate transport of nos mRNA in proximal dendrites. PMID: 24027279
  7. Mei-P26 associates with Bam, Bgcn and Sxl and nanos mRNA during early germline stem cell development, suggesting that this protein helps to repress the translation of nanos mRNA. PMID: 23526974
  8. nanos is a critical regulatory target of polar granule component in primordial germ cells. PMID: 23173091
  9. results suggest that Sxl enables the switch from germline stem cell to committed daughter cell by posttranscriptional down-regulation of nanos expression PMID: 22645327
  10. show that, during late oogenesis, nos translation is also repressed post-initiation and provide evidence that Glo mediates this event PMID: 21460235
  11. The minimal promoter (from -108 to +20) of Nanos is sufficient in the germline stem cells for the green fluorescent protein expression in transgenic Drosophila. PMID: 20026207
  12. Ectopic expression of the Translational Control Element of nos indicates that a common translational control mechanism functions in axial patterning and neuroendocrine signaling in Drosophila. PMID: 12091303
  13. removing Nanos from either germline stem cells (GSCs) or their precursors, PGCs, causes both cell types to differentiate into germline cysts; Nanos is essential for both establishing and maintaining GSCs by preventing their precocious entry into oogenesis PMID: 14976263
  14. Nos establishes germ-line/soma dichotomy and is also required to maintain germ-line fate. PMID: 15240884
  15. analysis of a translational control element in the nanos mRNA PMID: 15525666
  16. Nanos downregulates transcription and modulates CTD phosphorylation during Drosophila development PMID: 15817222
  17. The nanos translational control element represses translation in somatic cells by a Bearded box-like motif. PMID: 15936341
  18. Required to regulate the formation of the germ cell-specific spectrosome, further supporting a role for nos in promoting germ cell identity. PMID: 16028275
  19. Smaug response elements contained in the nanos 3'-untranslated region are necessary and sufficient to induce deadenylation PMID: 16793774
  20. Rumpelstiltskin regulates anterior-posterior axis patterning by functioning as a direct-acting nanos mRNA localization factor. PMID: 18234721
  21. Localization and translational control of nos mRNA are essential for dendritic arborization neuron morphogenesis. PMID: 18472422
  22. These observations suggest that Bam-Bgcn act together to antagonize Nos expression; thus, derepressing cystoblast-promoting factors; findings emphasize the importance of translational repression in balancing stem cell self-renewal and differentiation. PMID: 19470484
