Target Background

Function
Single-stranded telomeric DNA-binding protein that is required to protect the 3'-end telomeric overhang. It binds the consensus sequence 5'-GGTTAC-3'. Regulates telomerase and telomere length.
Gene References into Functions
  1. Thus, our results suggest that pik1(+) is required for the maintenance of circular chromosomes. PMID: 29410177
  2. The Pot1 DNA-binding domain is capable of binding DNA in at least two structurally distinct binding modes. PMID: 25074378
  3. Pot1 promiscuity is implemented through binding modes that thermodynamically compensate for base-substitutions through alternate stacking interactions and hydrogen-bonding networks. PMID: 23201273
  4. Results indicate an unanticipated ssDNA-binding mode in which two SpPot1 molecules bind an oligonucleotide containing two telomeric repeats. PMID: 21911358
  5. The high affinity conserved DNA-binding activity of Pot1 protein is conferred by a DNA-binding domain consisting of two subdomains. PMID: 21815629
  6. The Pot1 (protection of telomeres) protein prevents access of telomerase to the 3'-ends of telomeres and protects telomeric DNA against exonucleolytic degradation. [pot1] PMID: 15637058
  7. Pot1(1-389) presents a new model for studying and understanding the ssDNA-binding activity of S. pombe Pot1 and illustrates the malleability of both the ssDNA ligand and the oligosaccharidce-binding fold in the recognition and binding of telomeric ssDNA. PMID: 16842820
  8. Considering that the antiparallel tetraplex inhibits telomerase-mediated telomere elongation, we conclude that the ability of Pot1 to unfold the antiparallel tetraplex is required for telomerase-mediated telomere regulation. PMID: 17158862
  9. Characterization of the thermodynamics and accommodation of both cognate and noncognate telomeric single-stranded DNA (ssDNA) sequences by Pot1pN, an autonomous ssDNA-binding domain (residues 1-187) found in full-length S. pombe Pot1. PMID: 18355038
  10. study shows Tpz1 forms a complex with Pot1; Tpz1 binds to Ccq1 & Poz1 which protect telomeres redundantly & regulate telomerase in positive & negative manners; thus, Pot1-Tpz1 complex accomplishes its functions by recruiting effector molecules Ccq1 & Poz1 PMID: 18535244
  11. model in which conformational selection and induced fit play important roles in the recognition of ssDNA by Pot1pN (residues 1-187 of full-length Pot1) PMID: 19232358
