Description

The production of polyclonal antibodies targeting SEC16 necessitates repetitive immunizations of a rabbit with recombinant Saccharomyces cerevisiae (strain ATCC 204508 / S288c) (Baker's yeast) SEC16 protein (280-430aa). Once an adequate antibody titer is achieved, the rabbit is bled, and the serum-derived antibodies are purified via affinity chromatography. Subsequent evaluation of the resulting SEC16 antibody's functionality occurs through ELISA and WB assays, affirming its ability to interact with Saccharomyces cerevisiae (strain ATCC 204508 / S288c) (Baker's yeast) SEC16 protein.

The Saccharomyces cerevisiae SEC16 is a crucial player in the early stages of the secretory pathway. Operating at endoplasmic reticulum exit sites (ERES), SEC16 serves as a scaffold protein, organizing and maintaining functional ERES, which are vital for vesicle formation during protein transport. Additionally, SEC16 is implicated in cellular responses to stress conditions affecting the ER, highlighting its role in maintaining cellular homeostasis under challenging circumstances.

Target Background

Function
Involved in the initiation of assembly of the COPII coat required for the formation of transport vesicles from the endoplasmic reticulum (ER) and the selection of cargo molecules. Also involved in autophagy.
Gene References into Functions
  1. the present study sheds light on both the structural and the regulatory functions of Sec16. Our data explain at least in part the mechanism for how Sec16 functions in relation to the COPII coat proteins in the ER. PMID: 22675024
Subcellular Location
Endoplasmic reticulum membrane; Peripheral membrane protein; Cytoplasmic side. Note=On the endoplasmic reticulum and on vesicles which bud from it.
Protein Families
SEC16 family
