Description

This human DKK3 ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of human DKK3 in serum, plasma, or tissue homogenates. It also uses the enzyme-substrate chromogenic reaction to visualize and analyze the analyte levels through the color intensity. The intensity of the colored product is in direct proportion to the DKK3 levels in the sample and is measured at 450 nm through a microplate reader.

DKK3 is a secreted glycoprotein that acts as a regulator of Wnt signaling in organogenesis and carcinogenesis. DKK3 behaves as an oncosuppressor protein by inducing apoptotic death in a variety of cancers, although it might favor cancer cell spreading by promoting angiogenesis. In addition to functioning in tumorigenesis, DKK3 also plays a role in tumor neovascularization, prostatic acinus formation, cardiac vascular remodeling, renal and cardiac fibrosis, and immunological activity. DKK3 also modulates basic cellular processes, such as cell proliferation, differentiation, and apoptotic death.

Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease.
