The human IL-23 ELISA Kit is engineered for accurate measurement of human IL-23 levels from samples including serum, plasma, or tissue homogenates. It uses the Sandwich-ELISA mechanism in combination with the enzyme-substrate chromogenic reaction to measure the IL-23 content in the sample. The color intensity is positively correlated with IL-23 content in the sample. This kit has been validated against standards of sensitivity, specificity, precision, linearity, recovery, and lot-to-lot consistency.

IL-23 is a heterodimeric cytokine consisting of an IL-12p40 subunit and an IL-23-specific p19 subunit. It is secreted by activated dendritic cells and macrophages and can specifically amplify and stabilize the proliferation of IL-17-secreting Th17 cells. The IL-23/Th17 pathway has been involved in the development of many autoimmune diseases, such as rheumatoid arthritis (RA), systemic lupus erythematosus, and psoriatic arthritis (PsA). IL-23 signals through the IL-23R in complex with IL-12Rβ1, inducing the generation of secreted mediators such as IL-17 and IL-22, in pro-inflammatory, IL-17-secreting TCRγδ (Tγδ17), and in Th17 cells. These pro-inflammatory mediators facilitate the rapid recruitment and activation of granulocytes and macrophages that eventually result in the development of clinical symptoms.

Three samples of known concentration were tested twenty times on one plate to assess.

To assess the linearity of the assay, samples were spiked with high concentrations of human IL-23 in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

I am interested in measuring cytokines in patients affected by autoimmune thyroid diseases. For instance, i found the kit

Can this kit be used for the same purpose in lab animals (mouse, rats), if so, we are also interested in measuring IL-21, IL-22, IL-17A, IL-17F, all relevant kits produced by the same company, can they also be used for the same intention in lab animals??

Thanks for your inquiry. For these cytokines IL-23, IL-21, IL-22, IL-17A, IL-17F,detection of cross-species is not advised,as it will be affected by the characteristics of antibodies and the matrix difference.
Regarding your research in measuring cytokines in patients affected by autoimmune thyroid diseases,we suggest you use human kits.While regarding your early laboratory research on lab animals,we suggest you use the corresponding lab animals kits.

Interleukin-23 (IL-23) is a heterodimeric cytokine composed of an IL-12p40 subunit (that is shared with IL-12 ) and the IL-23p19 subunit.
This kit detects IL23(it is compound).
Capture antibody is mouse monoclonal antibody and detection antibody is goat polyclonal antibody.
Standard is a dimer of recombinant proteins expressed by eukaryotic cells in vitro.

Is the 293 protein you are referring the IL-23 p19 subunit?
Because you just mention the immunogen is 293 cell-derived recombinant protein. Please clarify, what is the 293 cell-derived protein specifically? For example, is it the IL-23 p19 subunit?
It is good to know, if the antibodies bind to the p40 unit or the p19 subunit?

Thanks for your further inquiry. There are 2 antibodies in the kit, so there are 2 immunogens. They are designed against the 2 subunits of the compound.
Sorry we can not disclose which antibody is specific to which protein.
