To assess the linearity of the assay, samples were spiked with high concentrations of human GLP-1 in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay. | ||||||

CUSABIO's GLP-1 human ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human GLP-1 in serum, plasma, or tissue homogenates. This assay uses an antibody specific to human GLP-1 coated on a 96-well plate. GLP-1 present in a sample is bound to the wells by the immobilized antibody after adding the sample into the wells. Biotinylated GLP-1 antibody is added to the wells, forming an antibody-analyte-antibody complex. HRP-conjugated avidin is pipetted to the wells and binds to the complex. A TMB substrate solution is added to the wells and color develops in proportion to the amount of GLP-1 bound. The addition of Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm using a microplate reader.
GLP-1 is a gastrointestinal peptide that is produced by the enteroendocrine L cells and is released in response to meal intake. GLP-1 mainly functions to stimulate insulin secretion and to suppress food intake, gastric emptying, and glucagon secretion, thereby limiting postprandial glucose excursions. It is important for glucose homeostasis. It also inhibits gastrointestinal motility and secretion and thus acts as an enterogastrone and part of the "ileal brake" mechanism. GLP-1 stimulates the proliferation, differentiation, and neogenesis of β cells.

For the CSB-E08119h, Human glucagon-like peptide-1, GLP-1 ELISA Kit, may I ask how we suppose to collect a blood sample and do we need to add DPP-4 inhibitor in EDTA-tube or not?
In Cusabio's instruction, there is no word that explains that we need a DPP-4 inhibitor while collecting blood plasma samples. Do we really not need to add a DPP-4 inhibitor to our sample if we use the Cusabio ELISA kit?
Actually, most CUSABIO ELISA kits are used to detect natural samples. And we usually do not do special treatment such as adding DPP 4 inhibitor in the sample when we detect serum or plasma samples. Though some references state that DPP 4 inhibitor (Dipeptidyl peptidase 4 inhibitors) is one medicine for type 2 diabetes, which could inhibit the inactivation of glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP), and enhance the level of endogenous GLP-1 and GIP. But we did not ever try this kind of operation. Not sure what the result will be.
