The human progranulin (PGRN) ELISA Kit is used to quantitatively measure PGRN concentrations in human serum, plasma, or cell culture supernates. It performs well in important characteristics, including sensitivity, specificity, precision, recovery, linearity, and lot-to-lot consistency. This assay is based on the sandwich ELISA mechanism and enzyme-substrate chromogenic reaction. The solution color develops proportionally to the amount of PGRN in the sample. And the intensity of the color can be measured at 450 nm via a microplate reader.
PGRN is a secreted growth factor that is involved in the modulation of multiple processes through itself or its conversion to granulins, including cell proliferation, survival, inflammation, wound healing, and tumorigenesis. It plays a major role in regulating lysosomal function and microglial responses in the central nervous system (CNS). In the brain, PGRN is mainly expressed by neurons and microglia and is involved in neuronal survival, neurite outgrowth, and synaptogenesis. It is a key regulator of neuroinflammation. Heterozygous mutations in PGRN lead to PGRN haploinsufficiency, which is one of the main causes of frontotemporal lobar degeneration (FTLD) related to the accumulation of TDP-43 inclusions.
