Because IL-22 is produced primarily by Th17 cells, ILC3s, and γδ T cells and acts locally at epithelial surfaces, researchers often need to detect it across both systemic and tissue-level compartments within a single study.
Detection Range: The 15.6–1000 pg/mL range captures IL-22 concentrations typically observed in mouse serum during colitis, dermatitis, or infection models, where levels commonly reach tens to hundreds of pg/mL. Most inflamed tissue homogenates and stimulated splenocyte supernates fall within this window without requiring extensive dilution, though samples from acute DSS colitis models at peak inflammation may need dilution to stay on-curve.
Sensitivity: At 3.9 pg/mL, the kit detects IL-22 in naïve or mildly stimulated conditions where circulating levels are near-baseline — critical for establishing pre-disease time points or measuring output from low-frequency ILC3 populations in sorted cell cultures.
Sample Compatibility: Validated use across plasma, serum, tissue homogenates, and culture supernates allows researchers to correlate systemic IL-22 levels with local production in gut or skin tissue within the same experiment, while also linking findings to in vitro polarization assays.
Assay Efficiency: The 1–5 hour protocol using 50–100 µL sample volumes accommodates studies involving serial bleeds from individual mice and limited yields from lamina propria lymphocyte isolations, preserving material for parallel cytokine measurements.
