Description

The product CSB-MA186062I1m is a monoclonal antibody specifically against the G4S linker. A mouse is immunized multiple times with the immunogen G4S-BSA. After that, the G4S linker antibody-producing B cells are isolated from the immunized mouse's spleen and fused with the immortal myeloma cells to form the G4S linker antibody-producing hybridomas. The positive hybridomas are cultured for G4S linker antibody production. The G4S linker monoclonal antibody is isolated and purified from the cell culture supernatant through protein G affinity chromatography. Its purity exceeds 95%. This antibody has been validated in ELISA and FC. In a functional ELISA, three immobilized recombinant proteins containing G4S linker at 2 μg/mL can bind to this anti-G4S linker antibody, with the EC50 of 5.223 to 18.40 ng/mL, 17.89 to 45.51 ng/mL, and 2.705 to 8.752 ng/mL, respectively.

The G4S linker is a flexible synthetic peptide sequence, usually composed of Gly and Ser, with a repeating pentapeptide core sequence: Gly-Gly-Gly-Gly-Ser. This linker is mainly used to connect variant heavy chains (VH) and variant light chains (VL) when constructing scFvs and CARs. The G4S linker provides rotational freedom, which makes the structure of bivalent engineered antibody fragments more flexible, improving the affinity and specificity of antibody binding to its target antigen.

The G4S linker monoclonal antibody plays an important role in the preparation of CAR-T cells. The G4S linker-based antibodies can effectively select and enrich transgenic cells during the lentiviral packaging process. The G4S linker-specific monoclonal antibodies can also detect various types of CAR. These antibodies can detect different CAR NK cells in vitro, as well as in whole blood or patient-derived tumor spheroids, showing high specificity and sensitivity. G4S linker monoclonal antibodies can be compared with different staining methods by flow cytometry, providing researchers with an effective method to analyze the expression of CAR cells. In tumor immunotherapy, G4S linker monoclonal antibodies can help identify and enrich tumor cells expressing specific antigens.
