The G4S linker monoclonal antibody is produced by the G4S linker antibody-generating hybridomas. The hybridomas are formed by the fusion of the G4S linker antibody-producing B cells and myeloma cells. The G4S linker antibody-producing B cells are isolated from the mouse immunized with the immunogen G4S-BSA. The G4S linker monoclonal antibody undergoes protein G affinity chromatography purification. Its purity reaches up to 95%. It has been validated in ELISA and FC applications. In a functional ELISA, three immobilized recombinant proteins containing G4S linker at 2 μg/mL can bind to this anti-G4S linker antibody, with the EC50 of 10.03 to 17.44 ng/mL, 22.60 to 28.76 ng/mL, and 6.478 to 10.27 ng/mL, respectively.
The G4S (Gly-Gly-Gly-Gly-Ser) linker is a flexible peptide chain usually used to connect different domains of two proteins or antibodies. In scFv, the G4S linker connects the light chain (VL) and heavy chain (VH), allowing them to fold correctly and maintain antigen-binding activity. During the production of CAR-T cells, the G4S linker monoclonal antibody can monitor the expression level of CAR and ensure that the CAR construct is correct at different stages of production without protein degradation or misfolding. G4S linker monoclonal antibody can also help researchers understand the characteristics of G4S linker and its impact on CAR-T cell function, such as the effect of linker length and flexibility on antigen binding efficiency or signal transduction strength.
