  • IHC image of CSB-MA731035A0m diluted at 1:50 and staining in paraffin-embedded human skin tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

The cells were incubated in 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 1h at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/100 dilution for 30min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Description

The MERTNL monoclonal antibody is generated using the recombinant human MERTNL protein (46-311aa) as the immunogen to immunize mice. The B cells are isolated from the immunized mice and are fused with myeloma cells to produce hybridoma cells, which are screened to select the hybridoma cell line that secretes the MERTNL antibody. The MERTNL antibody-generating hybridoma cell line is injected into the mouse abdominal cavity. The MERTNL monoclonal antibody is purified and obtained from the mouse ascites using protein A affinity chromatography with a purity of over 95%. It is an unconjugated IgG2a antibody that recognizes human MERTNL protein and is suitable for use in ELISA, IHC and FC applications.

MERTNL is a protein that acts as a ligand for the MER receptor tyrosine kinase, which is a member of the TAM family of receptor tyrosine kinases that play important roles in the regulation of immune responses, inflammation, and homeostasis. MERTNL is primarily expressed in the placenta and has been implicated in regulating the development and function of the placenta.

Target Background

Function
Hormone induced following exercise or cold exposure that promotes energy expenditure. Induced either in the skeletal muscle after exercise or in adipose tissue following cold exposure and is present in the circulation. Able to stimulate energy expenditure associated with the browning of the white fat depots and improves glucose tolerance. Does not promote an increase in a thermogenic gene program via direct action on adipocytes, but acts by stimulating several immune cell subtypes to enter the adipose tissue and activate their prothermogenic actions. Stimulates an eosinophil-dependent increase in IL4 expression and promotes alternative activation of adipose tissue macrophages, which are required for the increased expression of the thermogenic and anti-inflammatory gene programs in fat. Required for some cold-induced thermogenic responses, suggesting a role in metabolic adaptations to cold temperatures.
