This Macaca fascicularis ZG16B recombinant protein was produced in Mammalian cell, where the gene sequence encoding Macaca fascicularis ZG16B (23-169aa) was expressed with the C-terminal 10xHis-tag. The purity of this ZG16B protein was greater than 90%. Its endotoxin content is less than 1.0 EU/ug as determined by the LAL method. The activity was validated. In a functional ELISA, immobilized Macaca fascicularis ZG16B at 2 μg/mL can bind Anti-ZG16B recombinant antibody (CSB-RA836195MA1HU) with the EC50 of 20.15-28.98 ng/mL.
ZG16B (zymogen granulin 16 homolog B), a pancreatic cancer up-regulated factor (PAUF), is known to bind carbohydrates and activate inner epithelial cells, angiogenesis and permeability. It is highly expressed in pancreatic cancer and can increase the adhesion between pancreatic cancer cells, promote angiogenesis and increase vascular permeability of pancreatic cancer by activating CXCR4, β-catenin, TPL2/MEK/ERK, FAK/Scr and other signaling pathways way to promote the migration, metastasis and progression of pancreatic cancer. At the same time, these signaling pathways are closely related to gemcitabine resistance. Therefore, we speculate that ZG16B is also involved in regulating the drug resistance of gemcitabine to pancreatic cancer cells. Therefore, in-depth research on it is beneficial to clarify the mechanism of pancreatic cancer chemotherapy resistance and provide new targets for the treatment of pancreatic cancer.
