Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Description

Legumain is a lysosomal cysteine protease with strict specificity for asparagine residues, making it a valuable tool for dissecting asparaginyl endopeptidase activity in tumor microenvironments and antigen processing pathways. This full-length construct (aa 18–435) demonstrates robust catalytic function with a specific activity exceeding 400 pmol/min/µg against the fluorogenic substrate Z-AAN-AMC, providing quantitative benchmarks for kinetic parameter analysis including Km, Vmax, and kcat/Km determinations. The C-terminal His tag preserves access to the active site, supporting inhibitor screening campaigns, IC50 determination, and mechanistic studies of drug candidates targeting legumain-dependent processes. Purity greater than 95% by SDS-PAGE and endotoxin levels below 1.0 EU/µg meet the quality thresholds commonly required for accurate enzymatic assays, substrate specificity profiling, and use as a positive control in enzyme-linked detection systems.

Has a strict specificity for hydrolysis of asparaginyl bonds. Can also cleave aspartyl bonds slowly, especially under acidic conditions. May be involved in the processing of proteins for MHC class II antigen presentation in the lysosomal/endosomal system. Required for normal lysosomal protein degradation in renal proximal tubules. Required for normal degradation of internalized EGFR. Plays a role in the regulation of cell proliferation via its role in EGFR degradation.
