  • IHC image of CSB-PA22859A0Rb diluted at 1:300 and staining in paraffin-embedded human ovarian cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Description

The product CSB-PA22859A0Rb is a rabbit polyclonal antibody raised against recombinant human FAN1 (167-373aa). This FAN1 antibody is purified by protein G and gets a purity of up to 95%. It can recognize and detect human FAN1 protein in ELISA, WB, and IHC applications.

The FAN1 protein acts as a nuclease enzyme that plays a critical role in the repair of DNA damage, especially the interstrand crosslinks (ICLs). It has also been implicated in other cellular processes, including telomere maintenance and genome stability. Dysregulation of FAN1 expression or function has been associated with various pathological conditions, including cancer, neurodegenerative diseases, and autoimmune disorders.

Target Background

Function
Nuclease required for the repair of DNA interstrand cross-links (ICL) recruited at sites of DNA damage by monoubiquitinated FANCD2. Specifically involved in repair of ICL-induced DNA breaks by being required for efficient homologous recombination, probably in the resolution of homologous recombination intermediates. Not involved in DNA double-strand breaks resection. Acts as a 5'-3' exonuclease that anchors at a cut end of DNA and cleaves DNA successively at every third nucleotide, allowing to excise an ICL from one strand through flanking incisions. Probably keeps excising with 3'-flap annealing until it reaches and unhooks the ICL. Acts at sites that have a 5'-terminal phosphate anchor at a nick or a 1- or 2-nucleotide flap and is augmented by a 3' flap. Also has endonuclease activity toward 5'-flaps.
