Target Background

Function
Acetylates the free alpha-amino group of cysteine S-conjugates to form mercapturic acids. This is the final step in a major route for detoxification of a wide variety of reactive electrophiles which starts with their incorporation into glutathione S-conjugates. The glutathione S-conjugates are then further processed into cysteine S-conjugates and finally mercapturic acids which are water soluble and can be readily excreted in urine or bile. Alternatively, may have a lysine N-acetyltransferase activity catalyzing peptidyl-lysine N6-acetylation of various proteins. Thereby, may regulate apoptosis through the acetylation and the regulation of the expression of PROM1. May also regulate amyloid beta-peptide secretion through acetylation of BACE1 and the regulation of its expression in neurons.
Gene References into Functions
  1. Findings indicate that the NAA pathway has a prominent role in promoting tumor growth and represents a valuable target for anticancer therapy. PMID: 26819345
  2. ATase1 and ATase2 are endoplasmic reticulum-based acetyltransferases that form homo- and heterodimers and associate with members of the oligosaccharyltransferase complex PMID: 25301944
  3. Biochemical inhibition of the acetyltransferases ATase1 and ATase2 reduces beta-secretase (BACE1) levels and Abeta generation. PMID: 22267734
  4. NAT8L is the NAA biosynthetic enzyme Asp-N-acetyltransferase and is highly specific for the acetylation of aspartate. PMID: 20385109
  5. NAT8 is the enzyme that acetylates cysteine S-conjugates to mercapturic acids PMID: 20392701
  6. we raise the hypothesis that the alternative SNP alleles of the NAT8 upstream region may have differential effect on gene expression PMID: 18402670
