The recombinant Clostridium perfringens putative N-acetylmannosamine-6-phosphate 2-epimerase (nanE) is produced in E. coli. The expression construct contains the DNA fragment encoding the full-length nanE protein comprising amino acids 1 to 221 and carrying a 6xHis-tag at the N-terminus. The tag is helpful for recombinant nanE purification and detection. The purity of the recombinant nanE protein is determined to be over 90% using SDS-PAGE analysis. On the gel, this protein migrates as a band with an approximate molecular weight of 31 kDa.
The nanE protein plays a crucial role as an enzyme in the conversion of N-acetylmannosamine-6-phosphate (ManNAc-6-P) to N-acetylglucosamine-6-phosphate (GlcNAc-6-P). This enzymatic activity is involved in the biosynthesis of N-acetylneuraminic acid (Neu5Ac), which is a vital component of various cellular structures such as glycoproteins, glycolipids, and polysaccharides.
