Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Mechanosensitive channel that opens in response to stretch forces in the membrane lipid bilayer. Forms a nonselective ion channel with a conductance of about 4 nanosiemens. Participates in the regulation of osmotic pressure changes within the cell. Opens at a pressure just below that which would cause cell disruption and death. The force required to trigger channel opening depends on the membrane lipids composition.
Gene References into Functions
  1. Because MscL opens to tension in the lipid bilayer, requiring no other proteins or ligands, it could be developed into a general noninvasive sonogenetic tool to manipulate the activities of neurons or other cells and potential nanodevices. PMID: 29916253
  2. the amphipathic N-terminal helix of MscL acts as a crucial structural element during tension-induced gating, both stabilizing the closed state and coupling the channel to the membrane PMID: 27329693
  3. More importantly, the FE model indicates that to fully operate an MscL nanovalve by pulling directly on the N-terminus and bottom of TM1, gain-of-function (GOF) mutants (e.g., G22N MscL) would have to be employed rather than the WT MscL channel. PMID: 28859093
  4. Data suggest that dihydrostreptomycin binds to a specific site on mechanosensitive channel of large conductance (MscL) and modifies its conformation, thus allowing the passage of K+ and glutamate out of, and dihydrostreptomycin into, the cell. PMID: 27280286
  5. comprehensive screen has identified many residues that are critical for channel function as well as highlighted MscL domains and residues that undergo the most drastic environmental changes upon gating. PMID: 26368283
  6. Observe directly that the MscL channel opens via the helix-tilt model and the open pore reaches 2.8 nm in diameter. PMID: 24550255
  7. The crystal structure of the cytoplasmic domain of MscL contains canonical hydrophobic and atypical ionic interactions compared to previously characterized coiled coil structures. PMID: 24038743
  8. Results demonstrate the existence of oligomeric forms of MscL not yet observed by X-ray crystallography or other techniques and that in some cases a single type of MscL subunit can assemble as a mixture of oligomeric states. PMID: 21280123
  9. Data show that by using planar membrane bilayers MscL forms functional ion channels capable of ion transport. PMID: 21134371
  10. MscL does not need the insertase YidC for insertion in vitro PMID: 20888317
  11. Data show that introduction of a charge into pore lumen of MscL does indeed influence the permeation of charged molecules. PMID: 20930114
  12. the distribution of channels among the subconducting states of MscL is significantly less dependent on tension than the distribution between the closed and conducting states PMID: 15111402
  13. lipid effects altered activity through examined the contribution of specific headgroups by measuring their effect on the membrane tension required to gate the MscL channel PMID: 16142922
  14. Tyrosines and tryptophans that anchor both ends of the helices to membrane interfaces in many transmembrane proteins are not common in MscL and homologous mechanosensitive channels. PMID: 16156671
  15. YidC is essential for efficient membrane insertion of the MscL protein PMID: 17113597
  16. Pharmacological potential of MscL may involve discovery of new age antibiotics to combat multiple drug-resistant bacterial strains. PMID: 17350877
  17. model for structural transitions and residue-residue proximities that occur upon MscL gating. PMID: 19261722
