Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

 Q&A
Q:

I tested this protein in a functional assay, and found that it had low-levels of activity. Apparently, metal ions are necessary for the enzymatic function of this protein, and they were curious to know what metal ions (Zn ions, Fe ions, and Ni ions), if any, were specifically added to the culture medium. Could you please advise on which ones and at what concentrations?
Also, I was wondering if you had the ability to increase the concentration of metal ions, specifically 20 mM NiCl2, throughout the expression/purification process, to try and achieve higher activity. Would this be something your lab might be able to do for the customer?
I am fine with ordering the protein again, just as long as I can confirm to them that I can provide them with higher concentrations of metal ions during the expression/purification process. I also provided the article , which explains how to increase the metal ion concentrations to achieve higher activity.
Can you please let me know if any of this would be possible?

A:
Thank you very much for your feedback and the attached article. We could satisfy your requirement and increase the concentration of metal ions, specifically 20 mM NiCl2, to achieve higher activity, according to the method in the literature. Please kindly remark these requirements when sending your PO.
As we need to reproduce this protein, the lead time for the re-order will be 15-25 working days.
We hope you could share your re-testing result with us.

Target Background

Function
Removes the formyl group from the N-terminal Met of newly synthesized proteins. Requires at least a dipeptide for an efficient rate of reaction. N-terminal L-methionine is a prerequisite for activity but the enzyme has broad specificity at other positions.
Protein Families
Polypeptide deformylase family
