The recombinant human fibronectin type III domain-containing protein 5 (FNDC5) is produced in E.coli with the gene fragment corresponding to amino acids 32-143 of the human FNDC5. It is fused with a 6xHis-SUMO tag at the N-terminus. The 6xHis-SUMO motif is conducive to the generation of high-yield soluble, functional recombinant protein. The purity of this recombinant FNDC5 protein is greater than 90% measured by SDS-PAGE. It migrated to the molecular weight band of about 33 kDa on the gel. The target protein FNDC5 is a glycosylated type I membrane protein that produces a myokine irisin in response to exercise. Irisin travels to white adipose tissue and induces browning.
