Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The expression region of this recombinant Human DENR covers amino acids 2-198. The expected molecular weight for the DENR protein is calculated to be 38 kDa. The DENR protein was expressed in e.coli. The DENR gene fragment has been modified by fusing the N-terminal 6xHis-SUMO tag, providing convenience in detecting and purifying the recombinant DENR protein during the following stages.

Human density-regulated protein (DENR) is a conserved protein crucial for regulating translation. It plays a crucial role in the initiation and progression of protein synthesis. During translation, DENR forms a stable heterodimer with malignant T-cell-amplified sequence 1 (MCT-1), interacting with the 40S ribosomal subunit throughout initiation, reinitiation, and recycling stages. DENR, along with the oncoprotein MCT-1/MCTS1, supports unconventional translation initiation, facilitates translation reinitiation on specific mRNAs featuring short upstream reading frames, and plays a regulatory role in ribosome recycling.

Target Background

Function
May be involved in the translation of target mRNAs by scanning and recognition of the initiation codon. Involved in translation initiation; promotes recruitmnet of aminoacetyled initiator tRNA to P site of 40S ribosomes. Can promote release of deacylated tRNA and mRNA from recycled 40S subunits following ABCE1-mediated dissociation of post-termination ribosomal complexes into subunits. Plays a role in the modulation of the translational profile of a subset of cancer-related mRNAs when recruited to the translational initiation complex by the oncogene MCTS1.
Gene References into Functions
  1. findings elucidate how the DENR-MCT-1 dimer interacts with the ribosome and have functional implications for the mechanism of unconventional translation initiation and reinitiation. PMID: 28723557
  2. DENR binds to the P-site of the 40S ribosomal subunit and together with MCTS1 forms a tRNA binding surface and interfere with eIF1/eIF2/eIF3 binding, thus operating in ribosome recycling and translation re-initiation PMID: 28723557
  3. Reinitiation complexes involving initiation factors eIF2D, MCT-1, and DENR controls the translation of a large fraction of mammalian cellular mRNAs. PMID: 28732596
  4. both HuR and AUF1 bind to discrete regions of DENR/DRP mRNA and that AUF1 silencing increases DENR/DRP protein levels. PMID: 17878526
Protein Families
DENR family
Tissue Specificity
