Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Shelf Life

Description

VSIR functions as an inhibitory checkpoint receptor that dampens T-cell responses in the tumor microenvironment, making it a target of interest in immuno-oncology research. This mammalian-expressed construct spanning residues 33–194 captures the extracellular V-type immunoglobulin domain responsible for ligand engagement, and its native post-translational modifications support conformationally dependent antibody recognition, as demonstrated by an EC50 of 20.84–22.73 ng/mL in functional ELISA with an anti-VSIR recombinant antibody. The validated binding activity provides a suitable basis for therapeutic antibody epitope mapping, competitive inhibition screening, and affinity characterization by surface plasmon resonance or biolayer interferometry in checkpoint-focused drug discovery programs. With purity exceeding 95% by SDS-PAGE and endotoxin levels below 1.0 EU/μg, this protein aligns with standards expected in ligand-receptor interaction assays and blocking antibody validation workflows.
