The N-terminal 10xHis tag and C-terminal Myc tag was fused to the gene fragment corresponding to the 1-204aa of the human adenovirus C serotype 5 L3 protein and then was cloned into an expression vector. The expression vector was transformed into the E.coli for expression. The generated product was purified and separated to obtain the recombinant human adenovirus C serotype 5 L3 protein. Its purity is higher than 90%. This recombinant L3 protein showed an apparent molecular weight of about 30 kDa under SDS-PAGE condition.
Cleaves viral precursor proteins including pTP, pIIIa, pVI, pVII, pVIII, and pX could lead to the production of mature virions. Compared to the unmature virions, mature virions show a weakened organization, which further enhances the subsequent uncoating. The particle lacks infectivity and is unable to uncoat as there is no maturation. In the cytoplasm, L3Protease may participate in the cytoskeleton destruction and cleaves host cell cytoskeletal keratins K7 and K18.
