| Conjugate | Avi-tag Biotinylated E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag. |

From what I understood, the lab only expresses the expression region (89-117aa)? When produced in yeast, is it produced in Pichia pastoris? Also, this peptide is known to be a cyclotide, the circular structure is highly relevant for its bio-activity. The enzyme asparagine endoproteinase is essential for cyclization of the peptide. We would like to order this peptide to do some functional tests. I was wondering how are you making sure you are producing a mature circular peptide and that cyclization is taken place?
Yes, currentlly, we have only produced 89-117aa region from which the signal peptide and propeptide are removed.
Regarding the CSB-YP349763OAZ protein expressed by yeast, we use Pichia pastoris, and the commonly used strains are gs115 strain and X33 strain.
We are sorry that since it is a semi-custom product and hasn't been developed and produced yet, for the protein structure and activity, we can't test it, so we can't guarantee that it will meet the customer's expectations.
Looking at the structural studies in the literature, most of the sequences are constructed without signal peptide and N-terminal propeptide: https://www.uniprot.org/uniprotkb/P56254/entry#structure
In yeast expression, we use secretory vector, so the yeast protein is expressed in supernatant, which is very likely to be active. But customers still need to verify the activity themselves, because we do not guarantee the activity.
