Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Release of an N-terminal amino acid, cleave before cysteine, leucine as well as other amino acids. Degrades peptide hormones such as oxytocin, vasopressin and angiotensin III, and plays a role in maintaining homeostasis during pregnancy. May be involved in the inactivation of neuronal peptides in the brain. Cleaves Met-enkephalin and dynorphin. Binds angiotensin IV and may be the angiotensin IV receptor in the brain.
Gene References into Functions
  1. Significant role for IRAP in postnatal brain development and normal function of the hippocampus in adulthood. PMID: 27713012
  2. IRAP is more highly expressed in pro-inflammatory M1-activated macrophages. PMID: 24532823
  3. IRAP deficiency may lead to suppression of PAI-1 expression in adipocytes and upregulation of UCP-1-mediated thermogenesis in brown adipose tissue and increased energy expenditure to prevent the development of obesity. PMID: 25534853
  4. The current study demonstrates that deletion of the IRAP gene protects the brain from ischemic damage PMID: 21895534
  5. IRAP-deficient mice are less sensitive to the development of acute pentylenetetrazol-induced seizures, demonstrating that IRAP may be involved in generalized seizure generation. PMID: 21612947
  6. requirement of IRAP in cross-presentation extends to steady-state conventional dendritic cells (cDCs);data suggest increased recruitment of an IRAP(+)/Rab14(+) compartment to Ag-containing vesicles contributes to cross-presentation efficacy of CD8(+)cDCs PMID: 22238454
  7. These findings suggest that the IRAP binding protein, vimentin, plays an important role in retention of GLUT4 storage vesicles. PMID: 21216232
  8. IRAP functions in GLUT4 sorting from endosomes to GLUT4-specialized compartments. PMID: 20410133
  9. study shows the high-affinity binding site for angiotensin IV is absent in IRAP knockout mice brain sections in parallel with loss of IRAP immunostaining, providing irrefutable proof that IRAP is the specific high-affinity binding site for AT(4) ligands PMID: 19660563
  10. Data suggest that the physiological significance of circulating insulin-regulated aminopeptidase during human pregnancy cannot be addressed by investigations on mice. PMID: 19647771
  11. Initial characterization of knockout mice in which IRAP/P-LAP was deleted revealed that GLUT4 protein levels were dramatically decreased in all fat and muscle tissues. PMID: 12700100
  12. GLUT4 overexpression or deficiency affects the amount of other GLUT4-vesicle proteins including IRAP and VAMP2. PMID: 14985357
  13. GLUT4 and IRAP content of early endosome-derived sorting vesicles and of intracellular membrane vesicles are coordinately regulated. PMID: 15231875
  14. The sub-cellular localization of IRAP in neurons demonstrated in the present study bears striking parallels with distribution of IRAP in insulin responsive cells, where the enzyme plays a role in insulin-regulated glucose uptake. PMID: 17504262
  15. Changes in plasma and brain vasopressin levels in IRAP(-/-) mice suggest a significant role for IRAP in regulating vasopressin. PMID: 17684103
  16. GGA clathrin adaptors are not required for the re-entry of IRAP from the cell surface back into the IRC, whereas the Q-SNARE syntaxin 6 is required for this process PMID: 18388312
  17. Data suggest that modulation of glucose uptake by angiotensin IV and LVV-hemorphin 7 is dependent on colocalisation between IRAP and GLUT4, and confirm a role for IRAP and GLUT4 in activity-dependent glucose uptake in hippocampal neurones. PMID: 18702730
  18. lumenal interactions between Glut4 and IRAP play an important role in the assembly of the Glut4 storage vesicles PMID: 18713752
  19. Results suggest that placental leucine aminopeptidase might be involved in the increased oxytocin (OT) sensitivity just prior to onset of labor and also in the onset of labor by degradation of OT. PMID: 19249316
  20. study found a role for peptide trimming by IRAP in cross-presentation; in dendritic cells, IRAP was localized to a Rab14+ endosomal storage compartment in which it interacted with MHC class I molecules PMID: 19498108
  21. AngIV-AT4R signaling has a key role in fibrinolysis and the subsequent formation of arterial thrombosis after vascular injury. PMID: 19745198
