Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Function as phospholipase selective for phosphatidylcholine. May have a role in signal-induced cytoskeletal regulation and/or endocytosis.
Gene References into Functions

  2. PLD2 functions as a key mediator in the VEGF-mediated angiogenic functions of endothelial cells. PMID: 26818087
  3. PLD2 in neutrophils is essential for the pathogenesis of experimental sepsis PMID: 26282875
  4. The impact of polyunsaturated fatty acid (PUFA) supplementation on phospholipase D (PLD) trafficking and activity in mast cells was investigated. PMID: 23698760
  5. AQP3 has a pro-differentiative role in epidermal keratinocytes and PLD2 activity is necessary for this effect. PMID: 25233074
  6. Demonstrate a novel role for endothelial PLD2 in the survival and migration of ECs under hypoxia via the expression of hypoxia-inducible factor-1alpha and in pathological retinal angiogenesis and tumor angiogenesis in vivo. PMID: 24947526
  7. PLD2 is a unique GEF, with the PX being the major catalytic domain for its GEF activity, whereas the pleckstrin homology domain assists in the PX-mediated activity PMID: 23035122
  8. Data show that the Phospholipase D2 (PLD2)-GTPase Rac2 protein-protein interaction is sufficient for the guanine nucleotide exchange factor (GEF) function. PMID: 22106281
  9. study reports that a heterotrimeric protein complex exists between PLD2, Grb2, and WASp in vivo, which is integral to phagocytosis PMID: 21930784
  10. PLD2 positively affects Rac2 GTP loading activity of macrophages. PLD2 drives early chemotaxis. During early cell chemotaxis, PLD2 translocates to the cell membrane and facilitates lamellipodium extension. PMID: 21444720
  11. Walnut extract induced internalization of the LPS receptor, toll-like receptor 4, and that the anti-inflammatory effects of walnut were dependent on functional activation of phospholipase D2. PMID: 20213499
  12. Phospholipase D2-dependent inhibition of the nuclear hormone receptor PPARgamma by cyclic phosphatidic acid. PMID: 20705243
  13. Lipopolysaccharide-induced NO synthesis and iNOS expression were blocked by PLD2 siRNA, suggesting that lipopolysaccharide upregulates NO synthesis through PLD2. PMID: 19963059
  14. Protein kinase C in the PLD2 precipitates was constitutively activated. PMID: 12782152
  15. Elevating PLD activity in MCF-7 cells by transfection of mPLD2 led to rapamycin resistance; and inhibition of PLD activity in MDA-MB-231 cells increased rapamycin sensitivity. PMID: 12813467
  16. PLD2 was demonstrated to utilize physiological alcohol glycerol to form phosphatidylglycerol, which was elevated by extracellular Ca(2+) levels, but not 1,25-dihydroxyvitamin D(3). PMID: 14654225
  17. Phosphatidylglycerol, as a bioactive lipid, could potentially mediate the effects of the aquaporin 3-phospholipase D2 signaling module, with aquaporin 3 as a modulatory unit, in the regulation of keratinocyte function. PMID: 14675200
  18. PLD2 functions at the plasma membrane to facilitate endocytosis of the angiotensin II type 1 receptor. PMID: 14718562
  19. PLD2 isotype is expressed by mouse hippocampal astrocytes and is observed to increase dramatically in expression level along tracts of mossy fiber spouting, supporting the proposal that PLD plays a role in this process. PMID: 14999815
  20. PLD2 functions as a downstream signaling molecule of ERK to mediate the neuronal cell adhesion molecule L1-dependent neurite outgrowth of cerebellar granule neurons, a mechanism possibly related to alcohol-related neurodevelopmental disorders PMID: 15030398

  24. Taken together, these results suggest for the first time that hippocalcin can induce PLD2 expression and LPA potentiates hippocalcin-induced PLD2 expression, which is mediated by ERK activation. PMID: 16294323
  25. Data demonstrate that phosphatidic acid generated by phospholipase D2 plays an important role in cell survival during Fas-mediated apoptosis through the increased Bcl-2 and Bcl-xL protein levels which resulted from PLA(2) and AA-COX2 pathway. PMID: 17541981
