Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
MCH may act as a neurotransmitter or neuromodulator in a broad array of neuronal functions directed toward the regulation of goal-directed behavior, such as food intake, and general arousal.
Gene References into Functions
  1. our experiments reveal an insulin-dependent activation of MCH neurons in obesity, which contributes via distinct mechanisms to the manifestation of impaired locomotor activity and insulin resistance. PMID: 27926856
  2. Results indicate that olanzapine reduced accumbens nucleus GABAergic transmission and that MCH is necessary for the hypolocomotor effects of olanzapine PMID: 26092201
  3. The present work is the first study to systematically compare the effects of MCH signaling on behavioral and metabolic phenotypes.[meta-analysis] PMID: 24924345
  4. A functional cross-talk between MCH and IL-10 prevents resolution of inflammation in experimental colitis. PMID: 24556508
  5. acute activation of MCH neurons is sufficient, but not necessary, to trigger the transition from NREM to REM sleep and MCH neurons also play a role in the initiation and maintenance of NREM sleep. PMID: 24828644
  6. Sleep induction at a circadian time when all of the arousal neurons are active indicates that MCH stimulation can powerfully counteract the combined wake-promoting signal of the arousal neurons PMID: 23785141
  7. Rat prepro-MCH is processed into three neuropeptides: melanin-concentrating hormone, neuropeptide-glutamic acid-isoleucine (NEI), and Neuropeptide-glycine-glutamic acid (NGE). PMID: 1327720
  8. We conclude that MCH neurons regulate glucose tolerance through signaling molecules other than MCH. PMID: 23365238
  9. MCH was found to promote intestinal tumorigenesis at least in part by inhibiting epithelial cell apoptosis PMID: 22848656
  10. AgRP-expressing neurons play a critical role in mediating the metabolic syndrome and infertility of Lep(ob/ob) mice, whereas MCH-expressing neurons have only a minor role PMID: 22232663
  11. The hypothalamic inputs through MCH play an important role in regulating sensory integration from olfactory pathways. PMID: 21669232
  12. Studies suggest that rat and mouse MCH neurons are differentially involved in anatomical networks that control feeding and the sleep/wake cycle. PMID: 21103352
  13. Data suggest that the increased food intake and increased energy expenditure of Scd1-/- mice are independent of the neuropeptide melanin-concentrating hormone. PMID: 19883709
  14. importance of MCH in integration of energy homeostasis downstream of leptin and, in particular, the role of MCH in regulation of energy expenditure PMID: 12897241
  15. MCH triggers hyperphagia in BRS-3-deficient mice PMID: 14988239
  16. The hypothalamic arousal peptide hypocretin/orexin evoked a direct inward current and increased excitatory synaptic activity and spike frequency in the normally silent MCH neurons. PMID: 15157424
  17. enhanced wheel running activities in mice lacking either MCH or its cognate receptor PMID: 15544841
  18. These studies reinforce the important role of MCH in energy homeostasis and indicate that MCH is a plausible target for antiobesity therapy. PMID: 15731402
  19. MCH ablation improves the long-term outcome for several indicators of the aging process. PMID: 16443777
  20. We propose that alpha-DB plays a role in a structure or regulation mechanism unique to MCH-expressing neurons. PMID: 18314094
  21. MCH has a role in inflammatory processes in the intestine PMID: 18650383
  22. The results of this study support a role for MCH in vigilance state regulation in response to changes in energy homeostasis PMID: 18809470
  23. This study showed increased expression of MCH mRNA with fasting and in mice lacking leptin. It also demonstrated that administration of MCH in the lateral ventricle leads to increased feeding. PMID: 8637571
