Recombinant Neosartorya fumigata Hydrophobin (rodA) with an N-terminal 6xHis-B2M-tag is a full-length of mature protein expressed in E.coli. The sequence used to prepare this recombinant RodA protein corresponds to residues Leu19-Leu159 of Neosartorya fumigata RodA. Its purity is greater than 85% as determined by SDS-PAGE. A molecular mass band of approximately 30 kDa was visualized on the reducing SDS-PAGE gel. Applied for specific antibody synthesis, this recombinant rodA protein also may be used on Aspergillus fumigatus-related studies.
RodA is one of Neosartorya fumigata/Aspergillus fumigatus-expressed hydrophobins, which self-assemble spontaneously into the amphipathic layer at the hydrophilic-hydrophobic interfaces facilitating and supporting the fungal lifecycle at air-water and water-environmental interfaces. As the only needed hydrophobin in A. fumigatus, rodA determines the structure, permeability, hydrophobicity, and immunological inertia of the cell wall surface in conidia and is responsible for rodlet formation, conidiogenesis, resistance to physical insult or anionic dyes.
