To achieve the expression of the recombinant Pseudomonas putida (Arthrobacter siderocapsulatus) pht3 protein in e.coli cells, a DNA fragment encoding the Pseudomonas putida (Arthrobacter siderocapsulatus) pht3 protein (1-439aa) is inserted into a plasmid vector, which is then transferred to e.coli cells. After screening positive cells, they are cultured and induced to creat the pht3 protein. The protein features a N-terminal 10xHis tag and C-terminal Myc tag fusion. The recombinant Pseudomonas putida (Arthrobacter siderocapsulatus) pht3 protein is purified from the cell lysate through affinity purification. The purity of the obtained recombinant Pseudomonas putida (Arthrobacter siderocapsulatus) pht3 protein is greater than 85%, measured by SDS-PAGE.
