Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Mediates iodide uptake in the thyroid gland.
Gene References into Functions
  1. Nis expression is controlled by Txn/Ape1 through a TSH/selenium-dependent mechanism. PMID: 26650895
  2. excess iodide transcriptionally represses NIS gene expression through the impairment of Pax8 and p65 transcriptional activity. PMID: 26872612
  3. Report sodium binding date for NIS, where transporter activity is electrogenic, with a 2:1 Na(+)/I(-) transport stoichiometry. PMID: 26009546
  4. Although the affinity of NIS for I- is low, it increases when Na+ is bound. NIS takes advantage of the extracellular Na+ concentration and the pronounced increase in its own affinity for I- and for the second Na+ elicited by binding of the first. PMID: 24888603
  5. It is feasible to use lentiviral vectors to deliver sodium iodide symporter (NIS) gene for the non-invasive monitoring of bone marrowderived mesenchymal stem cell transplantation and survival in the infarcted myocardium in vivo. PMID: 25319483
  6. sodium/iodide symporter expression is inhibited by iodide excess in a process involving the PI3K/Akt signaling pathway PMID: 24424051
  7. Molecular imaging of luciferase signal and sodium-iodide symporter imaging may be useful for in vivo optimization of bioenergetics in transplanted cells. PMID: 23255420
  8. Overexpression of p38beta or Rac1 significantly enhanced (1.9- and 3.9-fold, respectively), the tRA-stimulated NIS expression in MCF-7 cells. PMID: 22157753
  9. This study found that KT5823 decreases iodide uptake within 0.5 h of treatment in trans-retinoic acid and hydrocortisone-treated MCF-7 breast cancer cells, but increases iodide uptake in thyroid cells. PMID: 21209020
  10. A physical interaction was observed between the NF-kappaB p65 subunit and paired box 8 (Pax8), which appears to be responsible for the synergic effect displayed by these transcription factors on NIS gene transcription. PMID: 20667985
  11. Data show for the first time that iodide regulates sodium-iodide symporter mRNA expression at the posttranscriptional level, providing a new mechanism by which iodide exerts its autoregulatory effect on thyroid. PMID: 20107044
  12. PI3K is involved in the IGF-I inhibition of TSH-induced sodium/iodide symporter gene expression PMID: 11818505
  13. an uncharged amino acid residue with a small side chain at position 395 is required for Na(+)/I(-) symporter activity PMID: 12145342
  14. sodium/iodide symporter gene is repressed by TGFbeta, which is regulated by interacting Pax8 and Smad3 PMID: 14623893
  15. The high degree of homology in the 5'-flanking region between mouse and rat NIS suggests that mNIS and rNIS share similar mechanisms for transcriptional regulation in the thyroid. PMID: 15009910
  16. Data show that tumor cells expressing the thyroidal sodium iodide symporter (NIS) retain iodide longer than thyroid cells due to a combination of slow efflux and reuptake. PMID: 15522214
  17. water transport across the membrane can be explained by cotransport of water in the membrane proteins and that intracellular unstirred layers effects are minute PMID: 16322051
  18. A significant proportion of human cholangiocarcinomas expresses membrane sodium iodide symporter, which may permit radioiodine therapy. Our data also suggest that (131)I acts on a crucial target for liver cancer development. PMID: 17408651
  19. MEK inhibition restores NIS protein levels and NIS-mediated radioiodide uptake activity in RET/PTC oncogene-transformed thyroid cells. PMID: 17639055
  20. that NIS protein levels and functional activity could be modulated by phosphorylation through distinct mechanisms. PMID: 17913707
  21. Gene transfer permits sequential real-time detection and quantification of reporter gene expression in the transplanted heart with micro-SPECT/CT imaging. PMID: 18165779
  22. The Na+/I- symporter mediates active iodide uptake in the intestine. PMID: 19052257
