Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

 Q&A
Q:

Please can you provide sequence, pricing, availability and tag information for all available sizes?
Have you previously attempted to remove the tag from this protein? I need a Rotavirus A VP6 recombinant protein without a His tag.
If this protein is not suitable, do you have one that would be more suited to our needs?
I am trying to develop an ELISA using an antibody I already made that has a His tag.

A:
Thanks for your inquiry. This protein is one of our best selling proteins, we will send you the COA of the previous Lot for your reference.
Recombinant Rotavirus A Intermediate capsid protein VP6
CSB-EP542165ROH >> E.coli
You can view the full sequence and tag information on this page.
Please remark your requirement for tag removal if you need the untagged protein or tag removal when placing the order.
Sorry, we haven't tried to remove the tag before. We can try enzyme digestion, but we can't guarantee 100% successfully.
The overall success rate of enzyme digestion data analysis is 75%-86%.
Not all protein tags can be removed as some proteins will be very unstable after tag removal.
If we succeed in removing the tag, we will charge for extra cost.
If we fail in removing the tag, we won’t charge for tag removal, and remark this information in datasheet as follows
“Note: The laboratory determined that the Tag on your protein could not be removed with standard laboratory procedures. Your protein is being supplied with the Tag intact.”
Generally, the delivery time will be extended for 3 days.
If we are failed in removing the tag, we will provide the fusion protein and won't charge any extra fees for tag removal.
Since you don't need His-Tag, will you accept the fusion protein with other tag such as gst-tag or some else if the tag removal is failed.
Q:

I am wondering if the protein - CSB-EP542165ROH -Rotavirus VP6 protein (His tag) -is folded similarly to how it would be on the surface of the virus. Alternatively, do antibodies from infected individuals bind to this protein?

A:
Very nice to receive your inquiry. The product details are confirmed as below.

The refolding form of the recombinant protein is not necessarily the same as the native form. We haven't tested yet. This protein we provide is full length of mature protein which is purified under mild conditions.
The antibodies from infected individuals can bind to this protein in theory.
CSB-EP542165ROH (E. coli) is expressed by Prokaryotic expression system. Generally, eukaryotic recombinant protein is closer to the native state. We have this protein expressed by Yeast system which have been delivered for many times. The following are the details of this eukaryotic recombinant protein.

Target Background

Function
Intermediate capsid protein that self assembles to form an icosahedral capsid with a T=13 symmetry, which consists of 230 trimers of VP6, with channels at each of its five-fold vertices. This capsid constitutes the middle concentric layer of the viral mature particle. The innermost VP2 capsid and the intermediate VP6 capsid remain intact following cell entry to protect the dsRNA from degradation and to prevent unfavorable antiviral responses in the host cell during all the replication cycle of the virus. Nascent transcripts are transcribed within the structural confines of this double-layered particle (DLP) and are extruded through the channels at the five-fold axes. VP6 is required for the transcription activity of the DLP.
