  • IHC image of CSB-RA261684A0HU diluted at 1:100 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
  • IHC image of CSB-RA261684A0HU diluted at 1:100 and staining in paraffin-embedded human breast cancer performed on a Leica BondTM system.
  • Overlay Peak curve showing Hela cells stained with CSB-RA261684A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Target Background

Function
Transcriptional repressor of genes that require a bHLH protein for their transcription. Plays an important role as neurogenesis negative regulator.
Gene References into Functions
  1. HES5 might contribute to the proliferation of non-small cell lung cancer by STAT3 signaling. PMID: 27878283
  2. that suppression of the HES5 leading to inhibition of proliferation may be one of the mechanisms against Hepatocellular carcinoma PMID: 26342546
  3. HES5 silencing is an early and recurrent change in prostate tumourigenesis PMID: 25560400
  4. these data identify HES5 as a key mediator of the Wnt-3a proneurogenic effect occurring independently of the classical Wnt/beta-catenin signaling cascade thus further deciphering crosstalk mechanisms of Wnt and Notch signaling pathways regulating cell fate PMID: 24548083
  5. the downstream Notch signalling effector HES5 directly represses transcription of the E3 ligase Fbw7beta. PMID: 23776410
  6. Notch3 and Hes5 are hypermethylated in human B cell acute lymphoblastic leukemia (ALL). PMID: 23637910
  7. Hes5 overactivation is associated with cell differentiation, thereby resulting in uterine carcinogenesis. PMID: 21495212
  8. In this study, HES 5 transcription factor was detected in the lining epithelium of human periapical cysts with limited inflammation, showing Notch pathway activation in those cells. PMID: 21238798
  9. The expression of HES5 in adenocarcinoma was significantly higher than those in adenoma and normal control PMID: 20091184
  10. For marking and purifying neural stem cells to ascertain whether differences exist, we generated transgenic mice using promoters from Hes genes (pHes1 or pHes5) to drive expression of destabilized enhanced green fluorescent protein. PMID: 16214363
  11. Notch protein binding to Hes5-GFP is extinguished fast and recovered slowly, whereas Hes1-GFP is inhibited late and recovered quickly PMID: 16365048
  12. The HLH gene HES5 on the other hand was only detected in chondrocytes. PMID: 17072841
