  • IHC image of CSB-RA156336A0HU diluted at 1:50 and staining in paraffin-embedded human lung cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.26% DAB.
  • Immunofluorescence staining of hela with CSB-RA156336A0HU at 1:20, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 500-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
  • Overlay Peak curve showing Hela cells surface stained with CSB-RA156336A0HU (red line) at 1:50. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.

Description

LGR6 (Leucine-rich repeat-containing G-protein coupled receptor 6) belongs to a family of receptors that play critical roles in stem cell biology and tissue homeostasis. As a marker of distinct stem cell populations in various tissues, LGR6 has emerged as an important target for researchers investigating regenerative processes, developmental biology, and cancer stem cell dynamics within signal transduction pathways.

This recombinant monoclonal antibody, clone 15G3, offers the reproducibility and consistency that demanding research applications require. Developed using recombinant technology with a sequence-defined approach, this antibody eliminates the lot-to-lot variability that can compromise longitudinal studies and multi-site collaborations. The rabbit IgG format, raised against a synthetic peptide derived from human LGR6, provides reliable specificity for your experimental workflows.

Validation studies demonstrate robust performance across multiple detection platforms. In immunohistochemistry, the antibody successfully identifies LGR6 in paraffin-embedded human lung cancer tissue using standard citrate buffer antigen retrieval, providing clear visualization with DAB chromogen detection. Immunofluorescence studies in HeLa cells reveal distinct staining patterns when counterstained with DAPI, enabling subcellular localization analysis. Flow cytometry validation confirms the antibody's utility for surface staining applications, with clear population separation demonstrated in HeLa cells compared to isotype controls.

The antibody arrives affinity-purified in a glycerol-containing buffer optimized for long-term stability at -20°C or -80°C. With validated applications spanning ELISA, IHC, IF, and flow cytometry, this antibody supports diverse experimental approaches for researchers exploring LGR6 biology in stem cell research, tissue regeneration studies, and oncology investigations where understanding receptor-mediated signaling is essential.

Target Background

Function
Receptor for R-spondins that potentiates the canonical Wnt signaling pathway and acts as a marker of multipotent stem cells in the epidermis. Upon binding to R-spondins (RSPO1, RSPO2, RSPO3 or RSPO4), associates with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase expression of target genes. In contrast to classical G-protein coupled receptors, does not activate heterotrimeric G-proteins to transduce the signal. May act as a tumor suppressor.
