  • IHC image of CSB-RA010380A17phHU diluted at 1:100 and staining in paraffin-embedded human lymph node tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

  • Immunofluorescence staining of MCF-7 cells with CSB-RA010380A17phHU at 1:56,counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4℃.The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG (H+L).

Description

Anti-phospho-Histone H1.4 (T17) antibody is a recombinant monoclonal antibody that recognizes the human Histone H1.4 phosphorylated at Thr17 residue. This phospho-Histone H1.4 (T17) antibody was drawn and isolated from the tissue culture supernatant (TCS) that cultivates the cell lines containing vectors of the human phospho-Histone H1.4 (T17) monoclonal antibody gene. It underwent affinity-chromatography purification. It is a rabbit IgG. And it can be used for ELISA, IHC, and IF testing with human samples.

The higher-order chromatin structure is maintained and established partially by the linker histone H1. H1.4, one of the 11 human H1 isoforms, is one of the most widely expressed somatic H1 variations, with significant levels of expression in many cells. Phosphorylation of H1 has been recognized as its most prominent modification. The antibodies against phospho-Histone H1.4 (T17) are helpful in the detection and localization of histone H1.4 phosphorylated at Thr17.

Function
Histone H1 protein binds to linker DNA between nucleosomes forming the macromolecular structure known as the chromatin fiber. Histones H1 are necessary for the condensation of nucleosome chains into higher-order structured fibers. Acts also as a regulator of individual gene transcription through chromatin remodeling, nucleosome spacing and DNA methylation.
