  • IHC image of CSB-RA878931MA1HU diluted at 1:100 and staining in paraffin-embedded human prostate cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.

Description

SIGLEC8, also known as sialic acid-binding Ig-like lectin 8, serves as a critical inhibitory receptor predominantly expressed on eosinophils and mast cells. This cell surface glycoprotein plays a pivotal role in regulating allergic inflammation and immune responses by recognizing sialylated glycan ligands, making it an attractive target for researchers investigating asthma, allergic diseases, and eosinophilic disorders. The selective expression pattern of SIGLEC8 on these specific immune cell populations has also positioned it as a promising therapeutic target in immunology research.

This recombinant monoclonal antibody, clone 18D13, offers researchers the reproducibility and consistency that only sequence-defined antibodies can provide. Generated against recombinant human SIGLEC8 protein and produced with human IgG1 framework, this antibody eliminates the lot-to-lot variability that can compromise longitudinal studies and multi-site collaborations. The affinity-chromatography purification ensures high purity suitable for demanding experimental workflows.

Validation in immunohistochemistry demonstrates reliable performance in formalin-fixed, paraffin-embedded human tissues. Testing in human prostate cancer tissue sections using citrate buffer antigen retrieval at pH 6.0 with dilutions of 1:100 yielded clear staining patterns when detected with HRP-conjugated secondary antibodies and DAB visualization. The antibody is also validated for ELISA applications, with recommended IHC dilutions ranging from 1:50 to 1:200 depending on tissue type and detection system sensitivity.

For investigators exploring signal transduction pathways in innate immunity or developing therapeutic strategies targeting eosinophil-mediated inflammation, this antibody provides a dependable tool for characterizing SIGLEC8 expression in human samples.

Target Background

Function
Putative adhesion molecule that mediates sialic-acid dependent binding to red blood cells. Preferentially binds to alpha-2,3-linked sialic acid. Also binds to alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. Recognizes simultaneously epitopes having a terminal N-acetylneuraminic acid (sialic acid) and an underlying 6-O-sulfated galactose. Preferentially binds to Gal-6-sulfated sialyl-Lewis X glycan epitopes.
