Description

Platelet basic protein (PPBP/PBP) is a CXC chemokine whose processed forms, including NAP-2, serve as potent neutrophil-activating signals central to innate immune recruitment and inflammation. This tag-free recombinant human PPBP fragment (residues 59–128) encompasses the mature NAP-2 sequence and demonstrates robust chemotactic activity on human peripheral blood neutrophils at 1.0–10.0 ng/ml, confirming signaling potency suitable for neutrophil chemotaxis and migration assays, CXCR2-mediated MAPK and NF-κB pathway dissection, and in vivo inflammation models. Endotoxin levels below 1.0 EU/μg minimize LPS-driven artifacts that can confound neutrophil activation readouts, while purity exceeding 97% by SDS-PAGE supports use as a reliable reference standard in cytokine detection assays or as a coating antigen for antibody development and ELISA validation. Produced in *E. coli*, this preparation aligns with standards expected in functional immune cell bioassays demanding both high specific activity and stringent endotoxin control.

LA-PF4 stimulates DNA synthesis, mitosis, glycolysis, intracellular cAMP accumulation, prostaglandin E2 secretion, and synthesis of hyaluronic acid and sulfated glycosaminoglycan. It also stimulates the formation and secretion of plasminogen activator by human synovial cells. NAP-2 is a ligand for CXCR1 and CXCR2, and NAP-2, NAP-2(73), NAP-2(74), NAP-2(1-66), and most potent NAP-2(1-63) are chemoattractants and activators for neutrophils. TC-1 and TC-2 are antibacterial proteins, in vitro released from activated platelet alpha-granules. CTAP-III(1-81) is more potent than CTAP-III desensitize chemokine-induced neutrophil activation.
