Description

The recombinant Escherichia coli (strain K12) ARCA protein is introduced into a rabbit, prompting the generation of antibodies that specifically target this antigen. Additional injections are administered to boost antibody production, and the resulting ARCA polyclonal antibodies are collected and purified from the rabbit's serum using protein A/G. Rigorous testing through ELISA and WB assays has validated the ARCA antibody's ability to specifically detect Escherichia coli (strain K12) ARCA protein.

The Escherichia coli (strain K12) ARCA protein mainly regulates gene expression. The ARCA protein helps regulate the expression of genes involved in capsule synthesis, allowing E. coli to adjust its capsule production in response to environmental conditions.

Target Background

Function
Member of the two-component regulatory system ArcB/ArcA. Represses a wide variety of aerobic enzymes under anaerobic conditions. Controls the resistance of E.coli to dyes; required for expression of the alkaline phosphatase and sex factor F genes; It also may be involved in the osmoregulation of envelope proteins. When activated by ArcB, it negatively regulates the expression of genes of aerobic function. Activates the transcription of the plfB operon by binding to its promoter.
Gene References into Functions
  1. The combined results indicate that ArcA controls metabolism, chemotaxis, and motility contributing to the pathogenicity of Avian pathogenic Escherichia coli PMID: 26099584
  2. We propose that this plasticity in ArcA binding site architecture provides both an efficient means of encoding binding sites for ArcA, sigma(70)-RNAP and perhaps other transcription factors PMID: 24146625
  3. The deletion of arcA results in a decrease of repression on transcription of tricarboxylic acid cycle genes under glucose abundant conditions, without significantly affecting the glyoxylate pathway activity. PMID: 21481254
  4. the activity of the regulator ArcA correlated with aerobiosis but not with the redox state of the ubiquinone pool, challenging the idea that ArcA activity is inhibited by oxidized ubiquinone. PMID: 21252224
  5. fadH promoter is regulated by three independent regulators, the fatty acid degradation FadR repressor, the oxygen-responsive ArcA-ArcB two-component system, and the cyclic AMP receptor protein-cyclic AMP. PMID: 20622065
  6. The ArcBA two-component system of Escherichia coli is regulated by the redox state of both the ubiquinone and the menaquinone pool. PMID: 19933363
  7. Knockout of ArcA-dependent redox regulation, however, increased the aerobic tricarboxylic acid (TCA) cycle activity by over 60%. PMID: 15838044
  8. ArcB/ArcA/RssB system is involved in sigmaS induction during entry into starvation conditions. PMID: 16291649
  9. must be regulatory factors other than ArcB interacting with ArcA to control flagella genes PMID: 17213678
  10. Activation of ArcA facilitates pIV-dependent induction of psp expression. PMID: 19682256
  11. These data show that the acetyl phosphate dependent ArcA phosphorylation route does not seem to play a significant role under physiological conditions. PMID: 19851741
