Description

In the production of the csgA polyclonal antibody, a recombinant Escherichia coli (strain K12) csgA protein segment (amino acids 45-134) is used as the immunogen to stimulate an antibody response in a rabbit. CUSABIO collects the rabbit serum and then purifies it using affinity chromatography to obtain the csgA polyclonal antibodies. The proficiency of the csgA antibody in detecting the Escherichia coli (strain K12) csgA protein is established through ELISA and WB assays, confirming its reliability for experimental applications.

The csgA protein in Escherichia coli (strain K12) is a major constituent of curli fibers, extracellular amyloid structures that serve crucial functions in bacterial physiology. CsgA is primarily involved in the formation of these curli fibers, which contribute to biofilm formation, adhesion to surfaces, and multicellular community development. The adhesion properties conferred by csgA and curli fibers enable E. coli to adhere to both biotic and abiotic surfaces, playing a role in colonization and persistence in various environments.

Target Background

Function
Curlin is the structural subunit of the curli fimbriae. Curli are coiled surface structures that assemble preferentially at growth temperatures below 37 degrees Celsius. Curli can bind to fibronectin.
Gene References into Functions
  1. CsgA deletion mutant lost its adherence to HTB-9 but continued to adhere to the HUVEC and Vero cells. PMID: 26991275
  2. A Monte Carlo study of the early steps of functional amyloid formation of bacterial biofilm, which is primarily formed by aggregates of the protein CsgA, has been presented. PMID: 26745180
  3. CsgA polymerization in vitro is preceded by the formation of thin needlelike protofibrils followed by aggregation of the amyloid fibrils. PMID: 21877724
  4. Data describe aspartic acid and glycine residues that function as "gatekeepers" to modulate CsgA polymerization efficiency and potential toxicity. PMID: 19966296
  5. At least three of five repeating units of CsgA are amyloidogenic, suggesting that covalent linkage of multiple amyloidogenic units facilitates efficient fiber formation. PMID: 17164238
