Target Background

Function
Catalytic component of the DNA polymerase epsilon complex which participates in chromosomal DNA replication. Required during synthesis of the leading DNA strands at the replication fork, binds at/or near replication origins and moves along DNA with the replication fork. Has 3'-5' proofreading exonuclease activity that corrects errors arising during DNA replication.
Gene References into Functions
  1. To dissect the Saccharomyces cerevisiae RNA polymerase II TL at individual-residue resolution, we quantitatively phenotyped nearly all TL single variants en masse. Our structural genetics approach incorporates structural and phenotypic data for high-resolution dissection of transcription mechanisms and their evolution, and is readily applicable to other essential yeast proteins. PMID: 27898685
  2. dNTP pool levels correlate with Pol epsilon mutator severity PMID: 25827226
  3. Authors report the presence of a [Fe-S] cluster directly within the active polymerase domain of Pol2 (residues 1-1187). PMID: 24144619
  4. Labeling of DNA polymerase epsilon catalytic subunit with the nick-containing DNA repair intermediate indicates that the DNA polymerase is involved in DNA repair in yeast PMID: 21601535
  5. show that subunits of DNA polymerase epsilon (Pol epsilon) are required but that Mrc1, a polymerase epsilon-associated checkpoint protein, is opposing senescence PMID: 21321081
  6. The active site location occupied by Met644 in Pol epsilon is a key determinant of DNA replication fidelity. PMID: 17452367
  7. RNAPII elongation is stimulated by H2B-K123 ubiquitination independent of H3-K4 methy PMID: 17543890
  8. findings reveal a pattern indicating that DNA polymerase epsilon participates in leading-strand DNA replication PMID: 17615360
  9. a mutation in the 3'-5' exonuclease domain of Pol2 dramatically reduces the frequency of end joins formed with initial 3' flaps. Thus, Pol2 performs a key 3' end-processing step in NHEJ. PMID: 18437220
