Target Background

Function
Protease that is part of the quality control system in the inner membrane of mitochondria. Activated in response to various mitochondrial stress, leading to the proteolytic cleavage of target proteins, such as OXA1 and COX1. Cleaves and thereby promotes the turnover of mistranslated or misfolded membrane proteins. Cleaves the misfolded multi-pass membrane protein OXA1. Involved in quality control of cytochrome oxidase assembly: mediates the cleavage of COX1 in cells lacking COA2. Required for the stability of the respiratory supercomplexes. Required for TOR signaling.
Gene References into Functions
  1. Study shows that in Saccharomyces cerevisiae, inactivation of the membrane-bound intramitochondrial quality control protease Oma1 interferes with oxidative-stress responses through enhanced production of reactive oxygen species during logarithmic growth and reduced stress signaling via the TORC1-Rim15-Msn2/Msn4 axis. PMID: 27325672
  2. yeast Oma1 is an important player in IM protein homeostasis and integrity by acting in concert with other intramitochondrial quality control components. PMID: 24648523
  3. Specific Cox1 proteolysis in coa2Delta cells arises from a combination of Oma1 activation and a susceptible conformation of Cox1. PMID: 22219186
Subcellular Location
Mitochondrion inner membrane; Single-pass membrane protein.
Protein Families
Peptidase M48 family
