Hi, I need to get Testosterone ELISA kits to test for the levels in flounder (fish) blood plasma.

This is a kit of small molecule and are of wide applicability on fishes.While we did not ever use them to validate on flounder samples.We suggest you do pre-testing first and take the actual measurement as the real result.

I want to know if your Estradiol and testosterone assays can be done in gonad tissues. If so, how can I homogenate those tissues?

May I know which species you need? human or mouse?
Here is the sample preparation method for tissue samples.
Tissue Homogenates 100mg tissue was rinsed with 1X PBS, homogenized in 1 ml of 1X PBS and stored overnight at -20°C. After two freeze-thaw cycles were performed to break the cell membranes, the homogenates were centrifuged for 5 minutes at 5000 x g, 2 - 8°C. The supernate was removed and assayed immediately. Alternatively, aliquot and store samples at -20°C or -80°C. Centrifuge the sample again after thawing before the assay. Avoid repeated freeze-thaw cycles.
After processing according to the above method, please be sure to do gradient pre-experiment.
